背景信息：

膜结合型转录因子蛋白酶位点 2 (MBTPS2)，也称作位点-2 蛋白酶 (S2P)，是高尔基体膜中的锌金属蛋白酶 (1,2,3)。它调节胆固醇代谢 (1,2) 和展开的叠蛋白质反应 (UPR) (3,4)。当细胞被剥夺胆固醇时，固醇调节元件结合蛋白 (SREBPs) 会从内质网 (ER) 移动至高尔基体，并且由位点-1 蛋白酶 (S1P) (5,6) 和位点-2 蛋白酶 (1,6) 依次剪切，以释放活性的氨基末端结构域。SREBP 的这些氨基末端结构域随后转移入胞核，以诱导胆固醇生物合成基因的表达。在 UPR 期间，激活的转录因子6 (ATF6) 从 ER 转运至高尔基体并且由 S1P 和 S2P 剪切以释放胞质片段。这个胞质片段迁移至胞核，并且激活 UPR 基因表达 (7)。
